FORMULATION, OPTIMIZATION AND CHARACTERIZATION OF ORNIDAZOLE NANOSTRUCTURED LIPID CARRIER GEL FOR TOPICAL DELIVERY

Dr. P. Ashok Kumar et al.
11/2/2026
pp. 191-209

The study aimed to formulate, optimize and characterize ornidazole NLCs using glyceryl monostearate (GMS), oleic acid and Tween 80 and to incorporate the optimized dispersion into a topical gel. NLCs were prepared by hot homogenization followed by ultrasonication. A three-factor, three-level Box–Behnken design comprising 17 runs was used to investigate GMS concentration, Tween 80 concentration and homogenization time. Particle size, polydispersity index (PDI), zeta potential, entrapment efficiency, drug content and in vitro drug release were evaluated. FTIR and SEM were used for physicochemical and morphological characterization. The NLC dispersion was incorporated into a Carbopol 934-based gel and evaluated for pH, spreadability, viscosity and stability. The optimized formulation (F13) had a particle size of 222.8 nm, PDI 0.289, zeta potential −26.7 mV, entrapment efficiency 83.6% and drug content 91.0%. The optimized formulation showed sustained drug release, with 82.3% cumulative drug release reported at 12 h in the response-optimization section. The optimized gel showed pH 5.73 ± 0.03, spreadability 6.28 ± 0.21 g·cm/s and viscosity 5080 ± 78 cP. FTIR analysis showed retention of characteristic ornidazole peaks without major drug–excipient interaction. The optimized formulation remained physically stable during the reported 60-day study. The study demonstrates the feasibility of developing an ornidazole-loaded NLC topical gel using a Box–Behnken design. The optimized formulation exhibited nanoscale particle size, acceptable PDI, measurable negative zeta potential, good entrapment and sustained drug release, supporting further investigation as a topical delivery system.

Design and In Vivo Evaluation of Acitretin Solid Lipid Nanoparticles Loaded Gel for Topical Delivery

Rajkumar Aland et al.
2/1/2019
pp. 1-13

Solid lipid nanoparticles (SLNs) are very potential formulations for topical delivery of antifungal drugs. The main objective for this investigation is to develop and optimize the SLNs of Acitretin for the effective topical delivery. Acitretin loaded SLNs were prepared by hot homogenization followed by the ultra sonication using Taguchi’s design and based on the results further investigation was made using central composite design (CCD). The lipid Dynasan-116, surfactant poloxomer-188 and co surfactant egg lecithin resulted in better percent drug loading and evaluated for particle size, zeta potential, TEM, drug entrapment efficiency, in vitro drug release and stability. All parameters were found to be in an acceptable range. In vitro drug release of optimized SLN formulation (F2) was found to be 95.63±1.52%, whereas pure drug release was 30.12 after 60 min. The optimized formulation was incorporated into the gel. The drug content of Acitretin gel formulation was found to 99.86 ± 0.012% and the diameter of gel formulation was 6.9 ± 0.021 cm and that of marketed gel was found to be 5.7 ± 0.06 cm, indicating better spreadability of SLN based gel formulation. The viscosity of gel formulation at 5 rpm was found to be 6.1 x 103± 0.4 x 103 cp. The release rate (flux) of Acitretin across the membrane and excised skin differs significantly. It is assumed that SLN based gel due to its appropriate physicochemical properties, high skin permeation ability, higher accumulative uptake in skin and improved skin tolerability may be more suitable as a novel regime for topical administration of Acitretin. This topically oriented SLN based gel formulation could be useful in providing site-specific dermal treatment of psoriasis with minimal drug systemic availability and high skin tolerability.

Formulation and Evaluation of Etoricoxib Topical Gel using Different Gelling Agent and Different Penetration Enhancer

Dinesh Chandra et al.
12/1/2013
pp. 1-13

The present investigation has been undertaken with aim to formulation and evaluation of etoricoxib gel with different gelling agent and different penetration enhancer. Etoricoxib is highly selective cyclooxygenase-2 (COX-2) inhibitor. In this present study gel with carbopol, HPMC and Na-CMC as gelling agent prepared with different penetration enhancer like propyl glycol, oleic acid. Menthol oil. Formulation were evaluated for pH, stability study, spreadibilty, extrudabilty, bioadhesive (ex-vivo), skin irritation, viscosity, appearance and In-vitro drug diffusion. The formulation of Etoricoxib topical gel was prepared using carbopol, HPMC and Na-cmc in three batches A, B and C. With the consideration of all formulation characteristic and parameter we selected batch-A formulation for further study of anti-inflammatory and anti-analgesic activity. It was concluded that the Etoricoxib gel formulation containing carbopol with increase concentration of propyl glycol 10% and 2% oleic acid (OA) was suitable for topical application and it shows comparable result with market product and shows much better result of formulation, anti-inflammatory and anti-analgesic activity.
